Syllabus

Course Objectives

Basic knowledge of solid phase synthesis strategies for peptides, oligosaccharides and nucleic acids with a critical approach on the appropriate synthetic choices aimed at lower environmental and economic impact on large and small scales.

Course Prerequisites

Basic knowledge of organic chemistry in accordance with the year of the course attended.

Teaching Methods

Theoretical lectures will be complemented by an in-depth examination of current literature, allowing students to explore the latest advancements in the field. Additionally, brief laboratory sessions will provide hands-on experience, comparing various peptide synthesis methods to evaluate their efficacy and efficiency. Students will have the opportunity to synthesize peptides, purify them, and conduct analyses as part of the practical component of the course.

Assessment Methods

The examination will be conducted through an oral test. Students will be asked to provide a concise critical analysis of a recent study from the literature, relating it to the foundational topics covered in the course. Additional questions pertaining to both the presented topic and other course material will be posed to ensure a comprehensive assessment. Final evaluation will consider the student's overall capacity to critically comprehend information on the topic and their ability to present it clearly, succinctly, and comprehensively.

The course will cover the fundamental principles necessary for conducting solid-phase synthesis of peptides and their modified versions, as well as oligosaccharides and nucleic acids. It will delve into the current synthesis strategies, purification techniques, and analysis methods for assessing the purity and conformational aspects of the final products, utilizing spectroscopic techniques. Specific topics include:

- Synthesis of peptides and peptidomimetics, including discussions on orthogonal protection methods and the careful selection of protective groups based on the amino acid utilized, as well as the formation of peptide bonds.
- Exploration of peptidomimetics development and the incorporation of non-natural amino acids.
- Solid-phase synthesis and purification of carbohydrates and their modified counterparts.
- Examination of the chemistry and structure of nucleic acids, alongside solid-phase synthesis techniques for oligodeoxyribo- and oligoribonucleotides.
- Insights into the utilization of oligonucleotides as therapeutic agents, particularly employing the antisense strategy.
- Detailed exploration of activation and coupling methods for all proposed syntheses, including considerations such as temperature control, inert environments, and the potential use of microwaves to enhance synthetic efficiency.
- Comprehensive coverage of appropriate purification methods for the synthesized products and techniques for conformational analysis.
