Obiettivi Formativi
Achievement of a good command of the main genetic mechanisms which are often conserved from microorganisms to higher eukaryotes.
Full understanding of the main gene expression regulatory systems, of signal transduction pathways and of the physiological signals that drive genetic regulation.
Understanding of the life cycle of a simple biological entity like a phage.

Prerequisiti
Familiarity with Genetics and Molecular Biology concepts is required.
Students are expected to possess a good knowledge of the types and functions of genetic materials and associated nucleic acids (mRNA, tRNA, plasmids...).
Students should also be familiar with the general mechanisms of replication, transcription and translation of the genetic information.

Metodi didattici
Lectures given with the help of power point slides and explanations on the board; wooclap platform will be used to verify the progress of the class. Slides and additional materials (articles, short videos, lectures from keynote scientists) will be available on the KIRO platform.
Verifica Apprendimento
Written test with 8 open questions aimed at verifying the understanding of the concepts.
Two questions are particularly important and require the accurate description of few key experiments that allow to understand the scientific approach to biological problems.
In answering the remaining questions, students must particularly focus on describing the mechanisms underlying each system described.

Plasmids: general properties, replication, partitioning and incompatibility. Conjugation: F plasmid, Hfr strains, Prime factors. From the discovery of conjugation to genetic maps. Genetics of bacteriophages. Lytic phages: developmental cycle, gene expression regulation and replication. Lysogenic phages: the Lambda paradigm. Genetic analysis with bacteria and phages. Benzer experiments with T4. Gene expression regulation: Promoters, RNA polymerase, sigma factors. Positive and negative transcriptional regulation; attenuation & antitermination. Regulons: Catabolite Repression, SOS, heat shock. Bacterial defense systems; CRISPR; Toxin-Antitoxin systems. Signal transduction in bacteria: two-component systems, quorum sensing, phosphorelay and sporulation.
