Syllabus

Course Objectives

The main objective of the course is to provide an in-depth knowledge of the methods of sexual reproduction, in particular of mammals, and of the cellular and molecular mechanisms underlying the development of the mammalian embryo from fertilization to implantation in the maternal uterus, of the progressive restriction of cellular stemness during differentiation processes and of how, in adult tissues, stem cells are regulated in their niches. The expected learning outcomes are the following: 1. knowledge of the development and differentiation methods of male and female gonads and gametes; 2. knowledge of the mechanisms of fertilization and peri-implantation embryonic development; 3. knowledge of the derivation of stem cells from mammalian embryos; 4. knowledge of the cellular and molecular characteristics that characterize the different types of stem cells; 5. knowledge of the principles of stem cell niches' organization; 6. knowledge of the specific language for the description of the cellular and molecular events underlying the reproduction and development events addressed during the course; 7. ability to communicate the scientific information acquired during the course and to process it through classroom discussions and written in the final exam

Course Prerequisites

Students are required to have adequate knowledge of the fundamentals of general biology, with particular reference to the structure and functioning of the cell, the ways in which cells interact and the organization of tissues.

Teaching Methods

The course is carried out through lectures and in-depth seminars using Power Point presentations. Lessons will available to students as pdf files. No educational or tutored seminars are planned.

Assessment Methods

Written exam answering two questions in full on the topics covered during the course. The time available is 75 minutes. Exams will be held in person using the Kirotesting platform, unless otherwise specified due to health emergencies. The outcome of the exam will be reported on the ESSE3 platform. Each student may request viewing his/her work, following an appointment writing to danilo.cimadomo@unipv.it

Embryonic genome activation. Imprinting. Genome equivalence. Assisted Reproduction Techniques. Cloning. Origin and specification of embryonic germ layers. Gastrulation (principles). BIOLOGY OF STEM CELLS Sources and molecular characteristics of stem cells. Origin of embryonic stem cells and their derivation methods. The pluripotency molecular core and its regulation. Differentiation of embryonic stem cells. Cell reprogramming and the induction of pluripotency. Characteristic and function of spermatogonia in the mammalian testis. Role of stem cells in adult tissues. The stem cell niche (principles).
