Obiettivi Formativi

The teaching of Advanced analytical and Bioanalytical chemistry aims at providing the knowledge and the methodological bases necessary for the development and the application of advanced analytical procedures for the qualitative and quantitative determination of target analytes in complex matrices, mainly biofluids. The student will acquire a broad spectrum of knowledge and skills in the bio-analytical field, which include:
- Knowledge of the fundamental principles and terminologies of the advanced analytical techniques
- Knowledge, planification and application of the methodologies for collection and storage of a representative sample for the analysis, the preparation of samples aimed at the determination of biomolecules in complex matrices
- Knowledge of the main features of the elaboration of data collected with advanced analytical techniques
- To know how to identify the analytical technique and methodology and/or the combination of advanced bio-analytical techniques most suitable for the quali- quantitative determination of the target analyte, with particular attention to the preservation of the analytes, to the specificity/selectivity/sensitivity of the analytical technique, and to the contamination
- Ability to evaluate and select the most suitable analytical technique for a target scenario
- Critical awareness of data quality and its diagnostic value when solving complex analytical problems
- Capability to identify errors and limitations through a rigorous review of the applied methodology

Prerequisiti

Basic knowledge of fundamental and of instrumental analytical chemistry acquired in teaching activities of the bachelor degree in chemistry (L27). Fundamental knowledge of introductory statistics (including mean, median, standard deviation, and related concepts).

Metodi didattici

This course blends lecture notes, slides, scientific publications as case studies provided and shared by the teacher. Some materials instrumental parts will be presented during the coursework for more practical knowledge. The instructor will also recommend conferences and seminars on topics related to the course, which may be worth up to 2 credits if discussed in the exam.

Verifica Apprendimento

The exam consists in an oral discussion. The exam consists of an initial section in which the student is asked to briefly and critically present a literature case study of its choice, relevant to the topics covered in the course. This is followed by in-depth questions on the presented case study and then on other topics discussed during the course.
During the oral interview, the following aspects will be considered:
- theoretical knowledge on advanced bio-analytical techniques and methodologies, both instrumental and pre-analytical
- theoretical-technical knowledge in the treatment and evaluation of analytical data
-Application of technical knowledge to solve an analytical problem.

• Advanced (27–30L): Demonstrates complete mastery of course topics. Independently adapts taught analytical strategies to novel applications and uses technical terminology accurately.
• Intermediate (23–26): Demonstrates a solid understanding of course topics. Clear presentation and logical organization of taught analytical strategies with competent use of technical terminology.
• Minimum (18–22): Demonstrates basic, superficial knowledge of course topics. Presentation is only partially structured, requiring occasional guidance, with inconsistent use of technical terminology.

Contenuti

The important aspects associated with the whole (bio)-analytical workflow, i.e., from sample preparation to data processing, analysis, and interpretation, will be presented, discussed, and illustrated. A special emphasis will be paid on sampling strategies, handling and preservation of the analytical sample. The most recent developments in the area of separation methods and mass spectrometry will be discussed, with examples from different areas as diverse as biomedical/pharmaceutical analysis, bio-nanomaterials. Analytical techniques for preparation and treatment of complex samples, e.g. advanced solid-phase extraction and liquid-liquid extractions (including microextractions), will be discussed with focus on green analytical chemistry principles.
The course will explore advanced instrumental analytical techniques such as gas chromatography (GC), liquid chromatography (HPLC), and advanced pseudo-chromatographic techniques applied to bioanalysis as well as advanced mass spectrometry approaches for qualitative and quantitative analysis. Microfluidic methods and novel mass spectrometry inlet systems and ionization modes will be discussed as key tools for portable analytical chemistry.
The subject will also provide you with an introduction to the -omics sciences, such as proteomics and metabolomics including targeted and untargeted strategies for biomolecules, and metallomics applications for metals and metalloids in biological systems.
